To improve specificity of oligonucleotide annealing, the synthesis step relies on a set of thermostable DNA ligase and polymerase enzymes. To date, several methods for gene synthesis have been described, such as the ligation of phosphorylated overlapping oligonucleotides, the Fok I method and a modified form of ligase chain reaction for gene synthesis. Additionally, several PCR assembly approaches have been described. They usually employ oligonucleotides of 40-50 nucleotides length that overlap each other. These oligonucleotides are designed to cover most of the sequence of both strands, and the full-length molecule is generated progressively by overlap extension (OE) PCR, thermodynamically balanced inside-out (TBIO) PCR or combined approaches. ==== Recurrent psychotic episodes ==== Placebo-controlled trials of both first- and second-generation antipsychotic drugs consistently demonstrate the superiority of active drugs over placebos in suppressing psychotic symptoms. A large meta-analysis of 38 trials of antipsychotic drugs in schizophrenia with acute psychotic episodes showed an effect size of about 0.5. There is little or no difference in efficacy among approved antipsychotic drugs, including both first- and second-generation agents. The efficacy of such drugs is suboptimal. Few patients achieve complete resolution of symptoms. Response rates, calculated using various cutoff values for symptom reduction, are low, and their interpretation is complicated by high placebo response rates and selective publication of clinical trial results.

== Production == Oleum is produced in the contact process, where sulfur is oxidized to sulfur trioxide which is subsequently dissolved in concentrated sulfuric acid. Sulfuric acid itself is regenerated by dilution of part of the oleum. The lead chamber process for sulfuric acid production was abandoned, partly because it could not produce sulfur trioxide or concentrated sulfuric acid directly due to corrosion of the lead, and absorption of NO2 gas. Until this process was made obsolete by the contact process, oleum had to be obtained through indirect methods. Historically, the biggest production of oleum came from the distillation of iron sulfates at Nordhausen, from which the historical name Nordhausen sulfuric acid is derived.

== Early life and education == Fred McLafferty was born in Evanston, Illinois in 1923, but attended grade school in Omaha, Nebraska, graduating from Omaha North High School in 1940. The urgent requirements of World War II accelerated his undergraduate studies at the University of Nebraska; he obtained his B.S. degree in 1943 and thereafter entered the US armed forces.
